Performance of a single-photon laser-scanning confocal fluorescence microscope
Abstract
We investigated the performance of a single-photon laser-scanning confocal fluorescence microscope (SP-LSCFM) constructed by attaching a beam-scanning optical setup to a conventional optical microscope. The optical sectioning capability of the SP-LSCFM is demonstrated by imaging 10-μm diameter FITC beads and an Alexa Fluor stained star-shaped alga, using the 488-nm line of an argon-ion laser. Results show successful high-contrast lateral images corresponding to the achievement of axial discrimination.
