Probing the attoliter focal volume of a high numerical aperture objective lens

Abstract

We demonstrate experimentally the possibility of probing the tight confocal volume of a high numerical aperture (NA = 1.3) objective lens using mirror samples and mounted gold nanobeads. This has allowed us to measure axial resolution down to 450 nm fullwidth-half-maximum (FWHM) which corresponds well to the theoretical value of 420 nm derived using vector theory. The developed protocol is a significant tool for engineering point spread functions and compensating aberrations in confocal imaging systems.
